DNAdamagesofthenormalcellsarecausedbyrepeatedcelldivisionandoxidativestress.CellularSenescence,astateofirreversIBLegrowtharrest,canbetriggeredinordertopreventDNA-damagedcellsfromgrowing.Senescence-associatedβ-galactosidase(SA-β-gal),whichisoverexpressedinsenescentcells,hasbeenwidelyusedasaMarkerofcellularsenescence.AlthoughX-galisawellknownreagenttodetectSA-β-gal,thesearefollowingdisadvantages:1)requirementoffixedcellsduetothepoorcell-permeABIlity,2)lowquantitativecapabilitybecauseofthedifficultyofthedeterminationofvisualdifferencebetweenstainedcellsandnotstainedcells,3)requirementofalongtimeofstaining.

CellularSenescenceDetectionKit-SPiDER-βGalallowstodetectSA-β-galwithhighsensitivityandeaseofuse.SPiDER-βGalisanewreagenttodetectβ-galactosidasewhichpossessesahighcell-permeabilityandahighretentivityinsidecells.SA-β-galaredetectedspecificallynotonlyinlivingcellsbutalsofixedcellsbyusingareagent(BafilomycinA1)toinhibitendogenousβ-galactosidaseactivity.Therefore,SPiDER-βGalcanbeappliedtoquantitativeanalysisbyflowcytometry.
